Saccharomyces cerevisiae yeast cells as a test system for assessing Zeocin toxicity

چکیده
Having unique genetic machinery and a high degree of conservation with higher eukaryotes, the yeast Saccharomyces cerevisiae is recognised as smart experimental system for studying modes chemical toxicity. The present study was undertaken to elucidate changes in intracellular redox homeostasis key macromolecule structure following exposure Zeocin. Cell populations logarithmic, quiescent (Q) non-quiescent (NQ) cells BY4741 were used model examine cytotoxic effect this radiomimetic. levels endogenous ROS, oxidized lipids, carbonylated proteins, glutathione analysed after treatment Zeocin (IC 50 ). An increase ROS production respectively increased oxidative stress detected all three types cell populations, highest being observed proliferating S. cells. response both stationary phase (Q NQ) included an overexpression glutathione. also showed very low DNA susceptibility concentration (100–300 µg/ml), presented no induced double-strand breaks (DSBs) macromolecule. Based on our research it could be concluded that cellular physiological state critical factor determining resistance environmental Q most robust.
